Science and Quality

Ashwagandha root extract (KSM-66®), (600 mg)

Ashwagandha (Withania somnifera), also known as Indian winter cherry or Indian ginseng, has been used in Ayurveda for thousands of years as a Rasayana (a Sanskrit word that means path of essence) referring to techniques that restore the body’s vitality. Ashwagandha is a renowned adaptogenic herb for its ability to help the body adapt and respond to the effects of stress.

Prajna Ayurveda’s ashwagandha is produced using “Green Chemistry” principles (no alcohol or chemical solvents) using a process that is consistent with that described in traditional Ayurveda texts. The KSM-66 extract is cultivated from whole roots organically grown in Rajasthan, India.

In a double-blind, randomized, placebo-controlled study 91 healthy perimenopausal women ages 45–60 years supplemented with 600 mg KSM-66 or placebo for eight weeks.1

  • At weeks 4 and 8, the KSM group had significantly greater reductions from baseline compared to the placebo group for:
    • Menopause Rating Scale (MRS) total score, and MRS somatic, psychological, urogenital subscores
      • KSM’s reduction in MRS total score was more than double that of placebo (23.5% vs. 10.9%) at week 8
    • Menopause-Specific Quality of Life Questionnaire (MENQoL) total score, and vasomotor subscore
    • Hot flash score (number of events per day)
      • KSM had an average reduction of 2.1 compared to 0.8 in the placebo group at week 8
    • At week 8, the KSM group had a significantly greater reduction from baseline compared to the placebo group for:
      • Follicle Stimulating Hormone (FSH)
    • At week 8, the KSM group had a significantly greater increase from baseline compared to the placebo group for:
      • Estradiol

In another double-blind, randomized, placebo-controlled trial, subjects taking KSM-66 saw a significant reduction in levels of cortisol—a primary stress hormone—after 60 days, compared to placebo. Subjects were then given well-recognized psychological questionnaire tests to measure the effect of KSM-66. Test scores showed a significant reduction in stress and a tremendous reduction in anxiousness.2

***In a prospective, double-blind, randomized placebo-controlled trial, 50 healthy athletic adults received either 600 mg daily of KSM-66 or placebo over 12 weeks to evaluate the efficacy of KSM-66 on cardiorespiratory endurance and quality of life.3 The KSM-66 group showed the following improvements:

  • Cardiorespiratory endurance (VO2 max measures translated from 20-meter shuttle run): VO2 max as a measurement determines an athlete's ability to take in, transport, and utilize oxygen.
  • KSM-66 resulted in a significant increase in maximum oxygen consumption (VO2 max) at Day 56 and Day 84 as compared to placebo.
  • KSM-66 was able to help support cardiorespiratory performance in healthy adults while helping to improve their perception of their quality of life.

In another double-blind, placebo-controlled trial, female participants looking to improve their sexual function, between the ages of 21-50 were given either a placebo or 600 mg of KSM-66 ashwagandha. After 8 weeks the KSM-66 ashwagandha was seen to improve sexual function and feelings to enhance sexual health.4

KSM-66® ashwagandha has also been clinically validated to provide support for men’s health by boosting muscle size, enhancing testosterone levels, and improving muscle recovery.5 Researchers believe ashwagandha works on oxidative and non-oxidative mechanisms to provide potential support for male sexual health. It may work by providing healthy antioxidant support and it may also work in other ways to support healthy cortisol levels.6

Plus, research shows that KSM-66® ashwagandha supports healthy weight management. In a double-blind, randomized, placebo-controlled trial, 50 subjects between the ages of 18–60 with a history of ongoing, stress were administered either a placebo or 600 mg daily of KSM-66® for 8 weeks. In conclusion this study suggests that ashwagandha root extract can be used for body weight management in adults under ongoing stress.7

Brahmi extract (Synapsa™), (320 mg)

Synapsa® is a patented and standardized extract of Bacopa monnieri, also known as Brahmi, which has been used in Ayurvedic tradition for centuries.

In a double-blind, placebo-controlled randomized trial, 62 healthy subjects between the ages of 18–60 years took Synapsa or placebo daily for 90 days. Supplementation with this patented brahmi extract resulted in significant improvement in working memory and rapid visual information processing.8

In another randomized, double-blind, placebo-controlled, crossover trial, 46 healthy subjects took of Synapsa or placebo daily for 12 weeks. Supplementation with Brahmi resulted in improvements in visual information processing speed, learning rate, memory consolidation, as well as reduction in occasional anxiousness.9

In a randomized controlled trial, 35 subjects between the ages of 55–70 years supplemented with Brahmi extract or placebo daily for 12 weeks. Those taking Brahmi demonstrated a significant increase in total memory score and logical memory sub-score compared to baseline.10

In an acute, double-blind, placebo-controlled crossover study, 24 healthy subjects between the ages of 18-56 years supplemented with Synapsa or placebo. Those taking Synapsa showed a significant improvement in their cognitive demand score, assessing mental function, versus placebo.11

Saffron Extract (stigma) (as Clear’ Saff), (30 mg)

Saffron is a Middle Eastern herb with strong antioxidant properties. Several studies show saffron’s support for mood health, starting in as little as one week. Plus, it’s shown to regulate stress levels and occasional anxiousness, and it may support better sleep quality.

In a double-blind placebo-controlled study 121 participants with low mood were given 28 mg of standardized saffron extract daily for 4 weeks. The saffron group had significant improvements compared to the placebo group for:

Tension, confusion, fatigue, and vigor and total mood disturbance.

Positive and Negative Affect Schedule (PANAS):

Score is based on ratings of positive effects such as alert, enthusiastic, and interested, as well as negative effects such as irritable, distressed, and nervous.12

In another double-blind trial, 30 adults with low mood were given 30 mg saffron or placebo for 6 weeks. The saffron group had greater improvement in mood health starting in just 1 week and they experienced less occasional anxiousness.13

The positive effects of saffron on sleep quality was shown in a double-blind placebo controlled study of 55 adults with poor sleep given either 28 mg of saffron extract or placebo for 28 days. The saffron group showed significant improvements in sleep quality, number of awakenings, and alertness upon awakening.14

LactoSpore® Bacillus coagulans (2 billion CFU)

LactoSpore is spore-forming, lactic acid-based probiotic containing the specific B. coagulans MTCC 5856 strain.

Two particular studies have shown positive effects with 2 billion CFU using LactoSpore B. coagulans. One randomized, double-blind, placebo-controlled trial with 31 participants with bowel issues showed that 2 billion CFU of LactoSpore® B. coagulans reduced GI discomfort compared to placebo over a 90-day study period with individuals also reporting an improvement in quality of life.15

In another randomized, double-blind, placebo-controlled trial with 40 individuals with bowel and mood issues, results showed that supplementation with LactoSpore B. coagulans led to an improvement in mood and support of mental health, a reduction in gastrointestinal discomfort, and an improvement in digestive quality of life.16

Vitamin B6 (as pyridoxal-5-phosphate) (15 mg), Folate (as L-methyltetrahydrofolate calcium) (680 mcg DFE/400 mcg)

This trio of active B vitamins is essential for supporting cognitive health, including memory and recall as well as normal homocysteine levels for optimal cognitive function and cardiovascular health.

In a randomized, double-blind, placebo-controlled trial, 900 adults with psychological distress were assigned either to a supplement group (400 mcg folic acid and 100 mcg vitamin B12) or placebo for 24 months. Supplementation resulted in significant improvements on immediate (p = 0.046) and delayed (p = 0.013) recall. A significant improvement in overall cognitive health (p=0.032) and homocysteine levels was also seen (p<0.001).17

Three hundred twenty-nine healthy subjects ages 60–74 years old, participated in a randomized, double-blind parallel trial on the effect of low dose vitamin Bs on the Framingham risk score. The vitamin B treatment included 400 mcg of folic acid, 2 mg vitamin B6, 10 mcg vitamin B12, and 50 mg vitamin C, taken daily for 12 months. The control group took only vitamin C supplementation for 12 months. By the end of the study, the results showed an improvement in homocysteine concentration and HDL for the vitamin B group. The Framingham risk score was increased in both groups. The improvements seen suggest that combination of folic acid, B12 and B6 lowers plasma homocysteine concentrations, promotes cardiovascular health, and supports healthy HDL levels and healthy homocysteine concentrations already within the normal range.18

Shankhapushpi Extract (Convolvulus pluricaulis, whole plant) (100 mg)

Shankhpushpi, which goes by the botanical name Convolvulus pluricaulis is a perennial plant native to India. It’s been used in Ayurvedic tradition for centuries for its various health-promoting benefits.
